Application rate trials with a nuclear polyhedrosis virus to control Spodoptera frugiperda (Smith) on maize Anais da SEB

Different concentrations of nuclear polyhedrosis virus were applied as aqueous suspensions using a tractor mounted or a backpack-manual sprayer to control Spodoptera frugiperda (Smith) larvae on maize (Zea mays L.). Larval mortality varied with the application equipment. For the tractor-mounted sprayer application of at least 2.5 x 10(12) P.I.B./ha was necessary to attain a control comparable to that obtained with the backpack-manual sprayer application (70.2%). For this application method it can be used the dose of 2.5 x 10(11) PI.B./ ha, butNPV persistence was very short. A greaterNPV persistence was obtained with doses over 1.25 x 10(12) P.I.B./ha (93.4% mortality). The occurrence of parasitoids in the experimental area increased larval mortality rate...

ABSTRACT The Brazilian Poplar Moth, Condylorrhiza vestigialis (Guenée), compromises the wood productivity of poplar trees (Populus sp.), mainly affecting the matchstick industry in southern Brazil. Considering the lack of information on rearing techniques for this insect, the objective of this study was to develop an artificial diet to rear C. vestigialis with biological characteristics similar to the wild insects. Characterization of a Chrysodeixis includens nucleopolyhedrovirus Isolate from Brazilian Cerrado and Assessment of its Co-Infection with Anticarsia gemmatalis multiple nucleopolyhedrovirus BABT

Abstract Chrysodeixis includens has become the major Lepidopteran pest of soybean crops, especially in the Brazilian Cerrado (savanna) region. A native isolate of Chrysodeixis includens nucleopolyhedrovirus (ChinNPV) from this region, Buritis, MG, was assessed for its biological and molecular features. In addition, in vitro co-infection with Anticarsia gemmatalis multiple nucleopolyhedrovirus (AgMNPV), another virus of an important soybean pest, was tested. The ChinNPV-Buritis isolate presented an average LC50 of 7,750 occlusion bodies (OBs)/ml of diet in C. includens larvae. The administration of baculoviruses to insects for bioassay purposes is carried out, in most cases, by contamination of food surfaces with a known amount of occlusion bodies (OBs). Since per os infection is the natural route of infection, occluded recombinant viruses containing crystal protein genes (cry1Ab and cry1Ac) from Bacillus thuringiensis were constructed for comparison with the baculovirus prototype Autographa californica nucleopolyhedrovirus (AcNPV). Differentiation of BP-type baculovirus strains using in situ hybridization ArchiMer

BP-type viruses infect wild and farmed shrimp species in the Americas and can cause high mortality in larval stages. Histologically, BP disease is characterized by the presence of occlusion bodies (OBs) in the nucleus of hepatopancreatic epithelial cells. By in situ hybridization using different molecular probes, we tested shrimp infected with BP-type viruses collected from several geographical areas. In one shrimp, a case of double infection by 2 different BP-type viruses was noted. This has not been previously reported. The molecular probes recognized only the Pacific strain of BP. Abstract: Wettable powder formulations by microencapsulation of viral occlusion bodies (OBs) of both Spodoptera frugiperda multiple nucleopolyhedrovirus from Colombia (SfCOL) and a genotypic variant (SfCOL-A) were evaluated for controlling the fall armyworm S. frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) in maize crops. Microencapsulation preserved OBs activity after three months of storage at 35ºC, where insecticidal activity loss was not greater than 12%. Additionally, the formulation protected the OBs against inactivation caused by UV-B radiation, retaining its insecticidal activity after 6 hours of UV laboratory exposure, in contrast to unformulated viral suspensions, which presented an Original Activity Remaining (OAR) between 12.1 and 50%. Enhanced activity of Anticarsia gemmatalis Hüb. (Lepidoptera: Noctuidae) nuclear polyhedrosis virus by boric acid in the laboratory Anais da SEB

Boric acid concentrations (0.02,0.03,0.045,0.067 and 0.101 g/100 ml of diet) were evaluated in combination with the Anticarsia gemmatalis Hüb. nuclear polyhedrosis virus (AgNPV) for enhanced virali activity against the insect. Seven days after inoculation, the median lethal concentration (LC50) was 1.52 x 10(5) for the AgNPV alone and 7.95 x 10² for the NPV mixed with 0.045g of boric acid/100 ml of diet. At subsequent evaluation dates (9,11 and 14 days after inoculation) LC50's for NPV+boric acid were ca. 4x lower than those observed for the NPV alone. The goal of this work was to assess the potential of lyophylized milk whey ultra filtration retentate (MWR) when associated to yeast extract (YE), glucose and Pluronic F68 (PF68) to partially replace fetal calf serum (FBS) in Spodoptera frugiperda Sf9 cells culture in Grace's medium, aiming AgMNPV baculovirus production.
